Indication of Demoxiclave 625

Demoxiclave 625 mg, also known as Co-amoxiclav, is indicated for the short-term treatment of bacterial infections at various sites in the body. 

• Upper respiratory tract infections (including ENT) such as tonsillitis, sinusitis, and otitis media.
• Lower respiratory tract infections including acute and chronic bronchitis, lobar and bronchopneumonia.
• Genito-urinary tract infections such as cystitis, urethritis, and pyelonephritis.
• Skin and soft tissue infections.
• Bone and joint infections such as osteomyelitis.
• Other infections like septic abortion, puerperal sepsis, intra-abdominal sepsis, etc.

Pharmacology

• Amoxicillin & Clavulanic Acid is an antibacterial combination consisting of the antibiotic Amoxicillin and the beta-lactamase inhibitor Clavulanic Acid.
• Amoxicillin has a broad spectrum of bactericidal activity against many Gram-positive & Gram-negative microorganisms.
• Clavulanic Acid inactivates beta-lactamase enzymes commonly found in microorganisms resistant to penicillins and cephalosporins, thus protecting Amoxicillin from degradation.

Dosage & Administration of Demoxiclave 625

• Oral dosage for adults and children over 12 years:
  • 375 mg tablet: One tablet three times a day. In severe infections, one 625 mg tablet three times a day or one 1 gm tablet twice a day.
• Dosage adjustment is necessary in patients with renal impairment or hepatic impairment.
• During pregnancy and lactation, use only if considered essential by the physician.
• Overdose problems are unlikely but may include gastrointestinal symptoms and disturbance of fluid and electrolyte balances.
• Co-amoxiclav may be removed from the circulation by hemodialysis.

Interaction

• Co-amoxiclav may prolong bleeding time and prothrombin time.
• It may reduce the efficacy of oral contraceptives.
• Concomitant use of allopurinol during treatment with amoxicillin can increase the likelihood of allergic skin reactions.

Contraindications

• History of Penicillin hypersensitivity.
• Contraindicated for patients with a previous history of Co-amoxiclav or Penicillin-associated cholestatic jaundice.

Pregnancy & Lactation

• Used for various bacterial infections, but caution in certain conditions like renal impairment.
• Take necessary precautions in bacterial infections, acute otitis media, endocarditis, etc.

Precautions & Warnings

• Use with caution in patients on anticoagulation therapy or with severe hepatic dysfunction.
• Dosage adjustment is required in patients with moderate or severe renal impairment.
• Adequate fluid intake and urinary output should be maintained to minimize the possibility of crystalluria during high-dose administration.
